Successful Validation in Forensic DNA Analysis
We are pleased to share the publication by Kristiaan van der Gaag from the Netherlands Forensic Institute in the journal Forensic Science International: Genetics: “Validation of the IDseek® OmniSTR™ global kit for autosomal STR profiling.”
This study highlights the Reverse Complement PCR (RC-PCR) method as an improved tool for routine massively parallel sequencing (MPS) of short tandem repeats (STRs).

Key points:
- The performance of OmniSTR™ with minimal amounts of DNA surpasses current MPS and CE methods;
- Sequence variations in SE33 result in significantly more variants compared to length variations;
- OmniSTR™'s tolerance to inhibitors such as humic and tannic acid is comparable to PPF6C, but improved for ethanol (EtOH);
- Automated analysis of reference samples achieves 99.94 % accuracy for 96.7 % of detected markers;
- Minor alleles down to 1.5 % relative to the dominant can be detected in mixtures with OmniSTR™.
